Zeiss Lattice Light Sheet (LLS) 

Instrument description

The Lattice Light-Sheet microscope is an advanced system for high-resolution live-cell imaging, minimizing phototoxicity and photobleaching while achieving near-isotropic resolution (~250 nm in X, Y, and Z).  It enables fast volumetric imaging (up to three volumes per second), allowing real-time observation of dynamic subcellular processes.  The system uses light-sheet illumination with a lattice of interfering Bessel beams, producing a thin and uniform sheet that improves axial resolution and reduces out-of-focus light.  An integrated incubation system controls temperature, humidity, and CO₂, enabling long-term, unsupervised live imaging experiments.

Access procedures

Internal users (CNR / affiliated institutions)

Instrument use must be booked in advance and approved by the instrument manager (elisa.deluca@cnr.it)

External users

External access is possible under the following agreements:

1.Scientific collaboration – use of the instrument within a joint research project or bilateral collaboration agreement.

2.Training – hands-on sessions supervised by qualified laboratory staff.

3.Use under technical assistance

4.Service – measurement and analysis performed by the facility staff on samples provided by the user

Elyra 7

Instrument description

The ELYRA 7 is a fluorescence microscope based on Structured Illumination Microscopy (SIM), including advanced Lattice SIM and SIM² reconstruction, achieving ~120 nm lateral resolution. It also functions as a conventional brightfield and fluorescence microscope and uses two sCMOS cameras for simultaneous high-speed acquisition.  An integrated incubation system controls temperature, humidity, and CO₂, enabling long-term, unsupervised live imaging experiments. The system integrates STORM and PALM techniques, also in TIRF mode, enabling super-resolution down to 20–30 nm through single-molecule localization. These approaches allow nanoscale analysis of biological structures, such as protein localization and membrane organization.
